Amanda doesn't die until some time after the events of ST: IV, which is the last time her character appears.
There's no Spock/ Uhura explicitly in TOS, although there are quite a few hints that they're close friends - they often seem to be exchanging glances, it's almost always Spock rather than anyone else who helps her up after the obligatory 'being thrown across the bridge during battle' scene, and they seem to be in the habit of playing music together. (There's also a really cringey scene in one of the first series where Uhura flirts with him in what, if meant seriously, is a shockingly desperate manner, but I try to pretend she was just winding him up...)
Nimoy and Nichols apparently wanted to suggest that their characters were, if not actually having an affair, then very close.
