E=mc^2 relates only to the rest energy of a particle, and not to the other energies (kinetic, potential, etc). A particle like a photon has energy but 0 rest mass, so you can have energy without mass.
I'm not sure how this comment is in anyway relevant to the topic at hand? Especially as we're not talking at the level of particles, but at molecules... ?
