I think the 'EEEEE' is an alarm going off, probably triggered by the arrival of Spike and the bugs. Notice that this noise stops in the third panel of the second page - the 'EEEE' only fills half the panel instead of all of it like in the previous ones - and Spike says, "Ah, better. Well, they know we're here."
Thank you, looks like you're right. It didn't occur to me; I thought alarm signals would look like "ALARM!!! ALARM!!!" or something like that. I'l still learning to read comics. :)
