If Dawn is still in trouble Buffy and Spike might end up in conflict (in the Deeper Well, Spike opposed to recalling Illyria and saving Fred if costs were of thousands of lives) Could be the first time when Spike will not support Buffy

Yes, that's pretty possible. Writers have to create a valid conflict between them to move their story forward.
