It's fairly simple, except not. Essentially, Tohno Shiki is your average everyday boy who enjoys being sarcastic. But when those glasses come off, it reveals a whole new world for Shiki. His glasses hold back the power of his Mystic Eyes of Death Perception.
Despite the freaky name, it's a rather BAMF ability. Essentially, it lets Shiki see lines and dots, which represent the target's fated destruction. Depending on how immortal and how long the target lived is how much he can actually see. Regardless, using any sharp object, he can cut these lines or pierce the dots. If he cuts a line, the corresponding body part is lopped off with little to no hope of regeneration. If he pierces a dot, the target's existence is destroyed. With the target being alive but not having an existence, a paradox is created which the world seeks to correct by killing the target. Both of these methods completely bypass immortality, regeneration, armor, and reincarnation. You can pretty much call Shiki the ultimate Anti-Hax weapon. Let it be noted that there are individuals who do not have a fated destruction or do not "exist", rendering them immune to Shiki's abilities. These individuals are few and far between but for some examples.
Arcuied: During a Full Moon, she can't die due to crazy plot hax. Her existence is rendered immortal because of the world's unwillingness to kill her off. Even without a full moon, she is capable of recovering because of her immense power. By reshaping her concept, she can regenerate wounds caused by Shiki.
Servants from F/SN: While they have lines and dots, Shiki will never be able to perceive them without dying from his brain exploding from overexertion. Therefore, in definition, they're rendered immune.
TATARI: Because its existence is theoretical, its existence can't be destroyed.
Also to be noted is that he moves like a human unless he is in a dangerous or stressed situation. If the proper conditions are met (dangerous situation or fighting an inhuman target) Shiki's Nanaya instincts begin to take over. With these, Shiki can move faster than the eye can follow, even capable of running up walls and along ceilings. His knife flashes out faster than even an inhuman eye can see, cutting up to seventeen times in one slice. His "instant kill zone" ranges around ten meters, the furthest he can move in one burst.
At this current point (1-25-2008), Shiki has been overusing his eyes to protect others or due to curses. His glasses have gone through two "boosts" due to his own eyes overpowering them. Unless something is solved on how to stop the power of his eyes from growing, insanity followed by death hovers at the edge of his conscious at all times.
