Oh yes, the virgin business is definitely true, Alucard actually asks Seras if she is a virgin before he shoots her through the lung. Integra actually mentions it as well.
It certainly applies to men as well as women, Intergra states clearly that a man or woman would have to be a virgin and bit by a vampire of the opposite sex. So it's not a physical thing per se. And I've always thought losing one's virginity with either gender would count though I can't back that up. Specifically what acts... well I won't go there, but regardless I think it's a safe guess Walter's out of luck at his age. But even in the anime, where the rule may not apply, he presumably would have become a vampire already if he were interested. (thinks of sick fanfic involving vampire Walter and Seras and quickly discards the idea)
Vampire Walter...Mwahaha... I actually thought he was a vampire, when I first saw the preview for Order 06: Dead Zone. With him, doing the teeth thing and going all badass on Yan and such *sparkle sparkle* XD

Virgins older than 12 must be hard to come by, these days ^^'
But if it's more of a physical problem, then it wouldn't matter anyway,
