Well, if you're into history the Rijksmuseum is probably a good bet, also the Anne Frank House (go as early as you can, the line gets ridiculous). Van Gogh Museum? You should do one of the canal boat tours, you'll be able to take some nice photos of the city and hear bits of history relating to things you go by. Walk by the flower market, because it's crazy awesome, and Dam Square because you can't be there and not go by Damn Square. Other than that, I'd say just walk the city, take in the sights, and go in any museums that strike your fancy!
If you are coming by car, you will be better off if you can park it outside of the canal area, as there is almost no parking whatsoever.
