At this point (where I'm evaluating Guitar Hero against Rock Band... not GH2 or GH3, but the first incarnation), I'd say that Rock Band is the better game. The reasons are simple:
1. You can download extra songs to your XBox 360's hard drive, which makes it more personal. This might be possible with the later versions of GH as well... I'm not sure.
2. Rock Band uses a bar descending the screen instead of GH's circle... it's easier for me to tell exactly when I'm supposed to "play" it... I tend to jump the gun on the circles. The circles are bigger, but the bars are easier for me.
3. Rock Band has the option of guitar or bass (both use the same controller, but that's a fairly minor detail), drums, or microphone. Guitar Hero is just guitar. The caveat to this is that GH4 will have drums, and the kit is laid out more realistically.
Rock Band guitar = DEFINITELY easier than GH guitar. I keep wanting B to get Rock Band for Wii now that it's out, but maybe it's better that neither of us has it till we pass the bar...
The downside to Rock Band is that the star power is almost impossible to engage without taking out an eye. Supposedly, it's easier with the aftermarket controller. My friend traded in the Strat... didn't like the flush buttons.

So yeah, I'd go Rock Band.
