This question is epic. The Simpsons is one of those shows that consistently makes me happy no matter how low I'm feeling. The newer episodes can go to hell, but anything before 2000 is guaranteed to make me happy and to give me enough fodder for 2 or 3 nerdy friendships. It's tough to pick a favourite character--they all have their golden moments.

"My son is also named Bort." is definitely one of the best lines of all times. The frosty tone is key. I'm also a fan of pretty much anything Agnes (Skinner's mother) says. "Oh, my name is Agnes and you know it's Agnes! It means lamb... lamb of God!"
I also love "Do you want to change your name to homer junior? The kids can call you Hoju!"
There are so many. They've permeated my life. Whenever I have to explain a bad mood, I always think of saying "The doll's trying to kill me and the toaster's been laughing at me." Also, I think of money being exchanged for goods and services just about whenever I buy anything.
My favourite episode of all time is probably the one with Lisa's substitute teacher from the 2nd season. It's so sweet AND hilarious. Plus, an uncredited appearance by Dustin Hoffman is always a bonus.
