So I first suggested George/Izzie, and I know there's a lot of hate for that, so if you don't want to go there, that's all good with me. My other two suggestions were Mark/Addison and Cristina/Burke. I'm kind of a stickler for in-character writing, so I'd love it if you picked the pairing with the characters you'd feel most comfortable with. I also don't mind if you'd like to stick to a purely-UST plot, like friendship with a hint of something more. That's awesome, too.
I love angst. A lot. It's probably sick how much I love it. So if you wrote angst, you'd probably get a big huge (internet)hug from me. But a dash of hopefulness in there is always nice. I don't mind the fluff, either, though, so if that's your thing, go for it!
So, basically, go with your gut. Whatever you feel will bring out your best work is what I'd love to read.
I know I've been almost no help here, but I hope you at least got a feel of what I'm like and what I'd like to read. ;)
