The girl at the start is the winner of the previous battle royale I guess she really has no point other than to show that only one person can survive and it can be the most unlikely person.
And at the end they are taking the weapons for protection because they are wanted for murder or thats what I got was going on but then again I could be wrong as well.
The girl at the beggining is from a previous Battle Royale and has nothing to do with Kitano's class. And at the end the reason why people are looking for Shuya and Noriko is because they escaped and broke the rules of Battle Royale, since the movie is science fiction and its showing Japan as a communist country the leaders of the country are probebly really pissed of at them for escaping their "Game" XD

As the other two said, girl with the teddy is the previous winner. The BR program was something that happened all the time, not just that once. In the novel, something like 50 9th grade classes were selected every year throughout Japan to participate in the program; the number in the movie was more like one a year. It was made out to be a huge deal, a great honor to take place in the game, so the end results were broadcast on the news.
If you watch the second movie (which I recommend you don't, because it's completely terrible), the teddy girl actually shows up again as part of Nanahara's group, who are labeled terrorists because they went against the government, broke the rules of the game.
I definitely recommend the book by Koushun Takami, it's even better than the movie and explains more about the way the government works and such.
