This is looking so creepy (in a good way). The red faces were unexpected but are entirely my favorite part, and the glowing eyes are very effective. I'm excited to see where this goes from here!
As for values, now that you have it digital, why not print out a small copy and attack it with color pencils or something, see how you like it either way?

I can visualize how the two options for the background will look, I'm just unsure of what effect I want. If the background is light at top and dark near the figures, then it might start to have a religious feeling... light from the heavens. But the plus side is that the glowing eyes will be accentuated. If the background is dark to light, it will have the more clinical and cool feel I want for the series, but the eyes will not pop as much.
