One of my absolute favorite moments between the two is in season 3. The episode is "Fresh Blood".

Sam: It's just, I wish you would drop the show and be my brother again, 'cause...just 'cause.

I love it because you expect Dean to agree, say he wishes he could be innocent... but no, he wishes it for his brother, instead. ♥
