Personality:
She is a woman not without her vices. Vanity is assuredly one of them, as she has maintained the image of a buxom blond twenty-something-year-old well into her fifties and is quite capable of changing her appearance at will in order to suit her purposes (usually to avoid debt-collectors). It often requires her assistant and apprentice Shizune to prompt her to do any work that is required of her, as she is much more prone to being distracted into a game of cards or dice, or to plop herself in front of a pachinko machine and whittle through all her/their money. Well-known for her love of drinks and gambling, if something can be bet on, chances are, Tsunade would be willing to take someone up on that bet. And she will probably lose; the woman's bad luck is about as legendary as her powers and her...uh, cup size. Infamously, she is known as the Legendary Sucker, and gamblers the world over know this; they're grinning and eager to take her money in a game of "chance," knowing she bets big and loses everything.
Of course, you had better hope she loses, because Tsunade's gambling win is a bad omen indeed.
Jiraiya initially teased her for her flat chest and earned himself many smacks upside the head. As she became bustier and "more attractive" to him, his flirtations, gropes, and attempts to solicit a date from her were equally greeted with the necessary punch. However, when he's not being a total lecher, the pair seem to get along well enough and enjoy each other's company out for drinks and reminiscing. Aside from Shizune, Jiraiya could well be her best friend. On the other hand, it is not entirely clear what Orochimaru and Tsunade's relationship might have been like, although he clearly sees nothing wrong with abusing her weaknesses, or being blunt and hurtful. Neither Tsunade nor Jiraiya seem surprised by his behavior, though, only somewhat saddened and, as the rift divides further and Orochimaru betrays his village, the sadness becomes pain and a degree of anger towards him.
With the deaths of her loved ones, Dan and Niwaki, Tsunade develops a severe case of hemophobia which, to this day, she has not yet be able to overcome. Given enough will and reason, perhaps...but not yet.
Probably from the experience of being a ninja, and from the losses she has suffered emotionally (not financially), Tsunade gives off the attitude of being rather harsh and sometimes cranky or short-tempered, often barking orders and throwing things at people to make them leave her be. She is not opposed to telling things bluntly and is not free with her compliments, but she does have a good heart and can be caring when the situation calls for it. At the time she is pulled from Tsunade is very resentful of the office of Hokage and all it stands for. Only a fool would be so proud to take that position and die for their village so stupidly. ...The death never seems to end.
Background:
Tsunade comes from a legacy of great ninja. She is the granddaughter of Senju Hashirama, the Shodai (first) Hokage and one of Konoha's founders (as such, sometimes hime or princess is attached to her name). She is grandniece of the Nindaime (second), pupil of the Sandaime (Third), and is herself supposed to be appointed the Godaime (fifth). Together with her former teammates, Jiraiya and Orochimaru, Tsunade is also one of the Legendary Sanin. The woman is renowned for her healing abilities the world over, and she is the strongest kunoichi ever produced by Konoha.
Naturally, the little girls admire her greatly.
Her credentials aside, Tsunade began her ninja career as a flat-chested young genin under the tutelage of Sarutobi Hiruzen and teamed with Jiraiya (a young pervert-in-training who enjoyed teasing her) and Orochimaru (a genius with a rather creepy penchant for experimenting on things and a desire to master every jutsu ever). She had a natural talent for medical jutsu, and chose to focus on that skill as Konoha entered one of its most devastating wars...
On her brother Niwaki's twelfth birthday, Tsunade presented him with their grandfather's necklace, in honor of Niwaki's wish to become Hokage, and gave him a kiss on the forehead for good luck. The next day, the boy was dead, his body mutilated beyond all recognition.
After having spent years of the war watching ninja after ninja die needlessly on the battlefield simply because no one had enough skill to care for wounds, Tsunade finally made a plea to the Hokage (her former sensei) that they arrange for a trained medical ninja to become a permanent fixture on each squad. Hiruzen disagreed, saying they were in the middle of a war and the enemy would not wait for these accommodations. But one man, Dan, spoke out for her, backing her idea.
Dan's dream was also to become Hokage and to protect the village and all in it. Tsunade fell in love with him, and bestowed her grandfather's necklace on him along with a kiss on the forehead.
...Despite Tsunade's best efforts to save him, Dan died on the battlefield not long after that. His death, along with the cumulative toll taken by the deaths of comrades, and of her brother, and Orochimaru's betrayal, destroyed much of Tsunade's faith in the village, and the ninja world. Disillusioned, she packed up her things and, with Dan's niece Shizune as her aide and apprentice, Tsunade left the village, her remaining teammate Jiraiya, and some of her most painful memories behind her for a life on the road.
She has be gambling and racking up a debt ever since.
...Flash-forward some years later.
Tsunade is now around fifty years old. She has arrived in Takzaku-Gai to bet and lose her suitcase full of money, when unbeknownst to her both of her former teammates (and their apprentices) are simultaneously seeking her out.
It is then that Tsunade lands triple sevens on the slots.
No good could come of such extraordinary luck.
Tsunade would have left the market-city just then, if only Shizune had not wanted to sightsee at the castle. It is there that Orochimaru and Kabuto, atop Manda the snake, find the two women and corner them.
No time is wasted before explaining what they are here for-only Tsunade has the power to fix Orochimaru's sealed chakra and useless arms, and he needs them in order to destroy the leaf village. The death of her former sensei at the hands of her former teammate comes as a bit of a shock and Tsunade wants nothing to do with the deal he has offered, at least not until he exposes his trump card and sweetens the deal: Just as Orochimaru brought back the first and second Hokage for the battle with the Third, he can also revive Dan and Niwaki...but only if Tsunade holds up her end of the bargain.
The offer is tempting, wrong as it is, and even though she would never enable Orochimaru to destroy the village, she is given a week to decide and to come forward, before Orochimaru and Kabuto will force her into it. Shizune is genuinely worried that Tsunade may take that offer...and rightly so. There are no two people Tsunade would rather have back than Dan and Niwaki.
Later that evening, the two women sit alone in a pub. By pure chance, or sheer dumb luck, Jiraiya and Naruto chose to scrounge for information at that exact same bar and ...lo and behold! There, sitting at a bench-seat and enjoying her sake, is exactly the blond Jiraiya's been looking for! It seems tonight is the night for reunions.
The former teammates chat, but she doesn't tell him about Orochimaru's proposal. Jiraiya, however, doesn't skirt around his issue and quickly tells her exactly why he has come for her: the elders have decided to name her as the fifth Hokage.
Tsunade declines. No one but a fool would risk his or her life so easily. Hokage's always die for their village.
Abilities/Additional Notes:
"Skilled" doesn't begin to cover her abilities in the medical field. Whether it is healing wounds, successfully performing otherwise hopeless surgeries, creating anti-toxins to lethal poisons, or leaving med-nin everywhere in awe and fear of her powers, there are few in the ninja world as skilled as Tsunade.
Creation Rebirth - using the chakra stored in the seal on her forehead, she can regenerate cells, even organs, in her body, but at great cost to her lifespan. She rarely uses it, therefore, except in dire circumstances.
Katsuyu - her summon, a giant white and blue slug, which can split into many smaller slugs to spread information to allies or to carry out healing over a battlefield when Tsunade herself cannot reach all the wounded.
Transformation - becoming an ageless, peerless beauty requires no fancy expensive anti-aging serums; instead, all it requires is a constant use of chakra to keep up the veil of transformation over her body. The effect is much steadier than any other transformation in use and rarely dispels, except when Tsunade reaches an extreme drop in chakra.
Taijutsu - fast, agile, and most importantly, strong, no one hits like Tsunade, who combines a lethal mix of raw power and chakra-infused punches for a truly devastating attack-or just to get her point across that no, Jiraiya, she is most definitely NOT interested. Now don't come back to the women's bath. EVER.

This is ridiculous. If anyone can hear over these things, I demand you tell me where Shizune is. That idiot Jiraiya is probably trying to woo her over to his side as we speak. The man never could take a hint, no matter how many times I smacked him upside the head.
[her voice lowers somewhat, dangerously] And if you have her, Orochimaru, I swear I'll kill you.

This wasn't Tanzaku, that was for certain, unless Orochimaru had become desperate and decided to wipe it out too, while he was at it. And Konoha's buildings were never this high, unless things really had changed in the past...however many years.

Long enough for the Elders to get some damn idea in their heads about her being good material for a Hokage after Hiruzen-sensei had gone off and stupidly let himself die for the village. Hokage was always Dan and Niwaki's dream, not hers. Why would the elders want her there anyway? She had no love for the village anymore.
Of course, she didn't want Orochimaru to destroy it, either.

Jiraiya was probably to blame for this, whatever this hallucination was, she thought grumpily. Maybe he, and Shizune, and that kid teamed up to slip a drug in her drink and drag her back to Konoha while she was out. That would be just like Shizune to go along with one of Jiraiya's idiotic ideas, all in the name of 'doing the right thing.' Tsunade had not missed how Shizune looked betrayed during the meeting with Orochimaru, as if she though Tsunade was going to accept the snake-nin's offer right then and there. The girl would do anything to stop her-she was as idealistic as Dan sometimes...just as foolish, too.
The building shook as she pounded her fist back once against the wall. A piece of debris and half the barbershop pole decorating the sign fell to ground not far off. Damn them, she thought bitterly. They were trying to get her to change her mind! Well she wasn't going to be their damn Hokage, no matter what Jiraiya said or did-even if he took her all the way back to Konoha while she was out cold. Even if Jiraiya could somehow bring back the dead too, her answer for him was still NO. Why couldn't he just leave her alone?
But if he groped her while she was out...!
