Purely theoritical, but there are two interesting, if somewhat rare things: Heteropaternal Superfecundation (that is, twins with different fathers) and Chimeras (two fertilized eggs merge in the womb into a single person with two sets of DNA). Now, both of these are rare occurrences in the first place, but there's really no reason why they shouldn't coincide...

Mitochondrial DNA wouldn't be from Nikola. It's only passed through women. If Ashley shared mitochondrial DNA with Nikola, she'd be sharing it with Nikola's mother.

So her parentage would be "Helen/John/Nikola's mum".
