Ezio is somewhat overconfident, and extremely playful despite his noble heritage. Even as an assassin he maintains his charasmatic charm, wooing various women he encounters on his missions and travels. Not everything is fun and games with Ezio Auditore though, as he can be quite serious when the time calls for it. He takes his missions with utmost seriousness in fact and dedicates himself fully to the cause of exacting revenge on those who killed his father and brothers. He also has a bit of a temper on him, unable to really control his anger until he was later trained in the assassin arts. He's extremely protective of his family members and close friends, and is obviously willing to help those in need.
As he aged, various things seemed to change about Ezio. He was calmer, more respectable toward his foes even with all they had done to him and his family. He realizes that killing Rodrigo, in the end, will not bring his loved ones back, and simply leaves him after a vicious beat down. That doesn't mean he lost all his aggression though, he is still a fiery spirit destined to carry on the legacy of his father before him. He has openly stated he does not enjoy the act of killing, even as an assassin.
Ezio has a tendency to draw people toward him, be it willing or not, and tends to make friends ( and enemies ) very easily. There are few he trusts though, and even fewer he would tell his secrets to. Those he loves know that deep down he is more than just a 'playboy' and a pretty face. He is a man on a mission, with fears and hopes and dreams just like anyone else, and there is more under the mask he wears than anyone could imagine.
During the course of the game Ezio meets many different people, and because of these people, his actions tend to change overall. When he learns of his father's true life as an assassin before his death, Ezio is enraged, unable to believe his uncle at first. It isn't until later when Vieri attacks that Ezio's outlook becomes strikingly different. He knows he has to fight, and he is more than willing to do so to protect his remaining family. It's from there on out that Ezio isn't just a rebellious playboy, but a determined, growing young man with a lot to learn about his destiny.
On the outside, he retains that playboy facade, covering up the insecurities he holds for the rest of his life. He is terrified that he cannot protect those that he holds dear, and therefore, tends to overcompensate by helping everyone and anyone he can if it is in his power to do so.
When finally inducted into the order, and told of his title as 'The Prophet' Ezio willingly accepts, if it will finally put and end to his quest for revenge, then he will accomplish it. Nothing had stopped him previously, and nothing would in the future. While by then his temper has diminished a good deal, he is till rather quick to anger, albeit a little less violent when he reacts. He holds more respect for those he kills, even if he doesn't enjoy the act itself.
He is more mature and level-headed, edging in becoming a master assassin and joining the ranks of his peers that have helped him through his years of training in secret. He still finds the need to protect those who cannot protect themselves, and holds all the friends he has made closer to his heart than ever before.
By the time of brotherhood Ezio is entirely different and yet very much the same. Over the years he has grown up, matured both mentally and physically. His anger has subsided, and he doesn't let his emotions get the best of him as much as they did when he was younger. In fact as a master assassin he can almost control it completely. almost. There were plenty of times when Ezio becomes openly frustrated, but it doesn't linger as long as it did previously. However he is still a very passionate man with many morals that he lives by. Even when on a mission he will go out of his way and divert from his original course to help someone in dire need along the way.
Ezio is still very flirty when it comes to women, that much, obviously, will never change. Another thing that hasn't changed is his sense of humour. Usually, when he likes someone he is likely to tease them as means of endearment. In some serious situations Ezio has been seen making odd, joking remarks at times. But that doesn't mean he does it all the time. No, Ezio does know when to be serious, and 9 times out of 10 he is. Ezio is also ridiculously stubborn still, refusing help after he was shot and immediately getting on a horse and traveling to Rome without getting medical assistance first.
Unfortunately at this point age is catching up to the assassin even if he is somewhat in denial over it. He speaks slower now, his voice is deeper and he acts like a true master assassin should, keeping those he needs under his wing and at his beck and call. He can rally troops with his speeches and he trusts in the brotherhood with his life. Such as with Machiavelli, he was determined to prove his innocence and loyalty to the brotherhood even when the man was seen talking to city guards. He is also dedicated to taking Rome back from Borgia rule, renovating everything he can to help his brothers and sisters along the way.
Ezio also still cares greatly for his friends and family, or what remains of them. He would always go to great lengths to help them, and often worries over them immensely. His worry is evident when Claudia and Maria show up in Rome and Ezio is visibly furious that they would risk coming there to help him. He begrudgingly accepted after a brief arguement with Claudia, but afterward there seemed to be a bit more tension than usual between the siblings. Ezio admittingly has kept his distance from his sister and mother through the years, and he obviously regrets not being for them at certain times.

I suppose it may have been stupid of me to assume I could make it to Rome after the villa was destroyed. The wound on my shoulder still pains me despite the medicine I received, but I'm refuse to give up so easily. The Borgia may have taken my home, the apple and my uncle's life, but I will not allow them the satisfaction of taking Rome as well.
The other assassins and I will rebuild and unite the citizens, Niccolo had the right idea when he suggested this. I will take the time to heal, but I cannot wait long. The apple is too precious and I need to find Caterina before they think to kill her as well.
There isn't much time, but as I did before I will do so now: I will take back what belongs to the order and destroy the corruption in this city. For the good of the people.

I thought this was all over, I thought that I was finished with my revenge. But it seems I was wrong again. I only wished to be with my family, or what remains of them, and enjoy the time I have with them. It is my responsibility and I must accept it. I will do this for my uncle and every other member of my family that once held the title of 'assassin'.
I may have spared Rodrigo once, but I won't make that mistake again. The Borgia will fall by my hand. I swear it.

Ezio sat atop the church's roof, reminiscing of all the times he had raced his older brother to the top of this very building, of when he captured feathers for Petruccio, or when he used it as a means to escape from the Pazzi guards hot on his heels. The moon was full, proving a wonderful light source for when he wanted to look out on his home. Or what was once his home. He recalled all the places he had been, all the things he had seen. And none were as beautiful as this.
As lovely as the Florentine skyline was, he could not enjoy it long. So he stood, albeit begrudgingly so, his gloved hand clutching the near invisible holds upon the side of the stone steeple beside him. He climbed with practiced ease, up to the top of the cross, taking the place of the lone bird that flew off into the night. While balancing with his arms stretched to either side, Ezio could see the golden outline of the man he was to kill that night.
His weigh shifted forward, sending the assassin down to the ground, a fall that would certainly kill him had he not landed safely in a stack of hay below. The rustling caused the surrounding guards to stop in their tracks, looking for the source of the subtle noise. Ezio was silent, waiting, a predator ready to pounce on its unsuspecting victims. When one of the guards rounded around the cart of hay, Ezio sprang into action, the blade on his wrist clicking out with a flick of his fingers and slamming down into the back of the guard's neck. In one quick motion Ezio had vanished back into the hay, dragging the dead man with him.
He did the same with the next guard, and the one after that. Littering the moonlit streets with bodies until he drove his blade right into his target's spine. The scream that followed was muffled when he clasped his free hand against the man's mouth, taking them both down so the man could give his last words.
And Ezio's only response was;
"Non abbiate paura del buio, ma il suo abbraccio di benvenuto. Requiescat in pace. ( do not fear the darkness, but welcome its embrace. Rest in peace. )" and retracted his bloodied blade, standing up and vanishing into the night in a flurry of robes.
