Apply for the position *and* others at the same time - if it's clear that you'll be leaving if you don't get the position, they'll be far more interested in giving you the position!

For that matter, I'd enquire sweetly as to what your supervisor's duties actually *are*.

I agree with all of the above. After all if you're doing one job application, you may as well do two or more at the same time.
