The earliest mention of "Shuffle Off to Buffalo" I can find is in 1931 released on an album by the...gah..something Sisters. I'll look it up again later. The musical was released in 1933. Even now, dances are named after songs. I think that's a tradition that goes back a long time. Shuffling off to buffalo is now considered an elementary dance step. Much like, say, doing the Twist.
I think it's pretty safe to assume the song came first.
