I've always wanted to learn to play guitar, but I've felt daunted by the prospect of doing so. Therefore, I have been teaching myself to play the ukulele in my spare time. I'm not so much kick-ass as tries-to-kick-and-lands-on-ass, but:

1. Make sure to use nylon strings instead of steel, to keep down on the hurtsies.

2. The most important tip for fretting is this: your fret hand should NOT BE SUPPORTING ANY OF THE WEIGHT OF THE INSTRUMENT. It's impossible to flex your fingers with the required freedom if you're simultaneously supporting the neck, even on something as light as a uke. (With a guitar you have to use a strap to accomplish this; with a uke, you press it against your body with your arms). It's amazing how much less flexibility you need to reach those chords when you have the freedom of moving your hand further around the neck so the fingers don't have to stretch so much. Which brings me to:

3. The thumb of your fret hand should be pressed against the back of the neck, not curled around the other side. The "grip the neck like a phallus" technique is reserved for unorthodox rock-star maneuvers where the thumb is used as a fifth fret-presser-downer (technical term).

Do not, under any circumstances, sprain your fret hand's wrist/carpal ligaments by, oh, I don't know, trying to hold 180lbs of dog on two leads in check with one hand. Just to pluck (ahem) a hypothetical example out of thin air. Of course, doing such a thing would also make typing a royal pain...
