He asks for ketamine because in a dream, he finds out that someone with his disorder/disability takes ketamine, goes into a coma, and comes out and then can walk.
Never having watched HOUSE, I'm not sure why he would want it. In real life, Ketamine is most commonly used in humans as an anaesthetic in patients with an unknown medical history, for minor surgery, or for treatment of migraine. The reason being that it suppresses heart rate and breathing much less than other drugs of type. Recently, a study of 17 patients led by Dr Carlos Zarate Jr. of the National Institue for Mental Health showed that ketamine significantly improved treatment-resistant major depression within two hours of injection. The improvement lasted up to one week after the single dose. This is still in the testing-experimental phase, of course, with work ongoing. Most interestingly, it has uses as a recreational drug with effects relatively similar to Phencyclidine and Dextromethorphan. (This drug was actually first formulated in 1962 to find a safer alternative to PCP, which was at the time being used as an anaesthetic.) It has also found uses in veternary medicine as well, for much the same purpose
