Just so I'm sure.. It's icons of one actor, five colored icons, not from the shows but from photo shoots they've done?

Your right about it, being about one actor of five icons and them being coloured.

However it can be of TV shows they'ev done they just have to be a Joss Whedon show. But if you wana use photoshoots you can use those photos even if it's from a non Joss Whedon show.

For example if i was doing James Marsters(Spike) i could icon him from Angel or Buffy using screencaps/episode stills and photoshoots. He was also in Smallville, but i wouldn't be able to icon him in that from screencaps/episode stills. But if he had any photoshoots promoting Smallville that would be okay to use.

Does it have to be just one character/actor or can it be more? For example, can I do a set of Angel/Cordy icons or do I need to choose between the two?

It should only be one actor/character so you'll need to choose, but a few(a max of 3 icons i'd say) of those may feature your actor/char with another person.
