Use, like the guy above said, Spybot search and destroy, as well as a program called Ad-aware by Lavasoft. In conjunction these two will keep your computer almost 100% free of spyware.

Just remember to click the update button on both programs before using them. Spyware changes constantly and Ad-aware in specific constantly releases new builds to combat the new versions.

Also, use Spybot's ability to immunize your computer. It will keep you from getting a lot of the bad stuff at all, instead of having to remove it later.
