I was only vaguely interested watching Lost's official pilot, but Part 2 is what made me a fan. They got me with the polar bear and the message. "They're all dead" still gets me after all this time.
The last 5 minutes of Doctor Who's "The Eleventh Hour" (which should count as a pilot because it was a new showrunner, new doctor, new companion... yes maybe please?)

Also Buffy Season 5 finale: Gift From the amazing opening sped-up montage of scenes from every single episode of the show up to that point, to the first scene which harkens back to "classic" Buffy, fighting one-on-one with a vampire while reflecting on how far she's come, to the climactic revelation that managed to tie all the threads of the season together in a hugely satisfying way, to "she saved the world, a lot.", it was all pretty much perfect. I remember running into high school science and 4 of us Buffy fans just congregated around one lab table squawking OHMYGOD at each other for the first 10 minutes.
Whilst looking back on the series as a whole and realising it's the weakest episode, I absolutely adored The Hour's opening episode. I saw it at a preview screening and I think there was something about watching it unfold on this huge screen that really made it work for me? Anyway, I put a picspam together for episode one: here
The premiere of White Collar!! I thought it was just going to be Matt Bomer parading around and looking pretty and, well, it is, but it's also fantastically written and I just can't stop watching.
